  2. 26 de dic. de 1997 · Specific binding of the plasmid-encoded protein, TrfA, and the Escherichia coli DnaA protein to the origin region (oriV) is required for the initiation of replication of the broad host range plasmid RK2.It has been shown that the DnaA protein which binds to DnaA boxes upstream of the TrfA-binding sites (iterons) cannot by itself form an open complex, but it enhances the formation of the open ...

Function. Required for initiation of plasmid DNA replication, along with host-derived DnaA and other host proteins. Both forms of the protein are capable of initiating plasmid replication in a number of Gram-negative bacteria. Binds to 8 17-base pair repeat sequences (iterons) in the RK2 minimal replication origin (oriV), opening the origin of ...

  4. 26 de nov. de 1998 · 1.. IntroductionPlasmid RK2 replicates at a defined copy number in a wide range group of Gram − bacteria. To initiate replication, the RK2-encoded TrfA protein binds specifically to five 17-bp repeats (termed iterons) in the plasmid minimal ori (Perri et al., 1991).TrfA also plays a key role in the maintenance of RK2 in the bacterial cell at a defined copy number.

  7. 17 de oct. de 2022 · TrfA protein. This family consists of several bacterial TrfA proteins. The trfA operon of broad-host-range IncP plasmids is essential to activate the origin of vegetative replication in diverse species. The trfA operon encodes two ORFs. The first ORF is highly conserved and encodes a putative single-stranded DNA binding protein (Ssb).
