To initiate replication, the RK2-encoded TrfA protein binds specifically to five 17-bp repeats (termed iterons) in the plasmid minimal ori (Perri et al., 1991).TrfA also plays a key role in the maintenance of RK2 in the bacterial cell at a defined copy number.

The trfA operon of broad-host-range IncP plasmids is essential to activate the origin of vegetative replication in diverse species. The trfA operon encodes two ORFs. The first ORF is highly conserved and encodes a putative single-stranded DNA binding protein (Ssb).
