Stanley Donen nació el 13 de abril de 1924 en Columbia, Carolina del Sur. Hijo de Mordecai Moses Donen y Helen Cohen. Tuvo una hermana menor: Carla. A pesar de haber nacido en una familia judía, Stanley Donen se convirtió en ateo en su juventud. Describió su infancia como solitaria e infeliz por pertenecer a la minoría judía de Columbia y sufrió ataques antisemitas en la escuela.
