Paul Theroux ist französisch-kanadisch-italienischer Abstammung. Er studierte an der University of Maine (1959–1960), an der University of Massachusetts ( Bachelor of Arts, 1963), und an der Syracuse University (1963). [1] Anschließend lebte er in Italien sowie in Malawi und Uganda, wo er als Lehrer arbeitete.

Paul Theroux si è sposato due volte: con Anne Castle dal 1967 al 1993 e successivamente con Sheila Donnelly (dal 18 novembre 1995). Attualmente vive alle Hawaii. Dal primo matrimonio ha avuto due figli Marcel Theroux e Louis Theroux, entrambi scrittori e presentatori televisivi, ed è lo zio dell'attore Justin Theroux.
