Mark Harmon est le fils du footballeur Tom Harmon et d' Elyse Knox, actrice de cinéma des années 1940, et fut le beau-frère du chanteur Ricky Nelson . Fan de baseball, il est en partie propriétaire d'une petite équipe, la « San Bernardino Spirit » qui apparaîtra dans son film Le Retour de Billy Wyatt.

Thomas Mark Harmon rođen je u Burbanku, u Kaliforniji, 1951. godine, u obitelji Toma i Elyse Knox. [1] Najmlađe je od njihovih troje djece. Otac mu je bio poznati igrač američkoga nogometa, te ratni heroj iz Drugoga svjetskog rata a majka mu je bila glumica i umjetnica. Harmon ima dvije starije sestre Kristin Nelson i Kelly Harmon.
