Tatăl lui Harmon, Tom Harmon este un cunoscut jucător de fotbal american iar mama sa actrița Elyse Knox.Harmon a studiat la University of California, unde a făcut parte între 1972 și 1973 din echipa de fotbal a universității. În anul 1973 i s-a decernat premiul National Football Foundation Award.
Mark Harmon est le fils du footballeur Tom Harmon et d' Elyse Knox, actrice de cinéma des années 1940, et fut le beau-frère du chanteur Ricky Nelson . Fan de baseball, il est en partie propriétaire d'une petite équipe, la « San Bernardino Spirit » qui apparaîtra dans son film Le Retour de Billy Wyatt.
