La USP surgió de la unión de la recién creada Facultad de Filosofía, Ciencias y Letras (FFCL) con las ya existentes Escuela Politécnica de São Paulo, Escuela Superior de Agricultura "Luiz de Queiroz", Facultad de Medicina, Facultad de Derecho y Facultad de Farmacia y Odontología.
Actualmente la USP es la mayor y más importante universidad pública brasileña, con 36 unidades de enseñanza e investigación distribuidas en seis campos: São Paulo, Bauru, Piracicaba, Pirassununga, Riberão Preto y São Carlos.
