Ryuzo is extremely pissed at the notion that he could become a samurai again. He wants nothing to do with it. Every time Jin tells him what Shimura can provide, he is barely receptive to it. Hell, he's incredibly apathetic when he learns Shimura is still alive, and this is when you very first speak to him.
At least, their involvement in the taking of Castle Kaneda. Ryuzo's reason for betraying Jin seems to have been born out of hunger and desperation. I guess I could see how their differences could also be a reason why, but it seems like Ryuzo's concerns for his men got the best of him, leading him to make extreme decisions.
