You can skip The Missing Pieces, but it's a nice watch. Fire Walk With Me is a movie, and was made to give the abrupt season 2 ending some closure. I wouldn't watch it without having sees S1 and S2. The whole premise and tag-line to Twin Peaks was "Who killed Laura Palmer?", but there's infinitely more to the show that just that.
