Zach Galifianakis urodził się 1 października 1969 roku w Wilkesboro w Północnej Karolinie (USA). Jego dziadkowie ze strony ojca wyemigrowali do Stanów Zjednoczonych z Grecji. Po ukończeniu Wilkes Central High School Galifianakis rozpoczął studia na Uniwersytecie Północnej Karoliny. Nie ukończył jednak uczelni.
